Several key points in the construction:
(1) Layout: independent buffer rooms should be set up in each functional room of the PCR laboratory, whether combined or distributed.
(2) Regional air flow: In order to avoid cross-contamination, the air flow in PCR laboratory must strictly follow a single direction, that is, only from reagent storage and preparation area → specimen preparation area → amplification reaction mixture preparation and amplification area → amplification product analysis area.
(3) Ventilation and air conditioning: in order to avoid cross pollution, the air in each functional room should not be mixed. The mixed PCR laboratory should adopt the fresh air direct current air conditioning system. When it is difficult to adopt the fresh air system, the air in each area can only be circulated in its own room.
(4) Biosafety: According to the Novel Coronavirus Laboratory Biosafety Guide (second edition), the 2019-nCoV pathogen is temporarily managed as class II pathogenic microorganisms, then the sample vegetation area should be negative pressure or enhanced P2 laboratory, and nucleic acid operation should be carried out in the biosafety cabinet.
